Azido-PEG3-acid (N3-PEG3-CH2CH2COOH) is a novel and potent PEG (polyethyleneglycol) analogue used as a PROTAC linker for the synthesis of BI-3663, BI-4216 and BI-0319. As a PEG-based linker, Azido-PEG3-acid is also used as a non-cleavable 3 unit PEG ADC linker for the synthesis of antibody-drug conjugates (ADCs). It is composed of an azido group, PEG spacer, and carboxylic acid. The terminal carboxylic acid can be reacted with primary amine groups in the presence of activators (e.g. EDC, or DCC) forming a stable amide bond. The azide group enables Click Chemistry. The hydrophilic PEG spacer increases solubility in aqueous media.
