背景信息:
Aurora A (AIK) 是一个细胞周期调控丝氨酸/苏氨酸蛋白激酶,在多种癌细胞系中过表达 (1-3)。Aurora A 在激酶激活环内的 Thr288 磷酸化后,导致蛋白活性大幅增加,并可能将蛋白在有丝分裂时靶标蛋白酶体降解 (4)。已发现密切相关激酶 Aurora B (AIM1) 在多个有丝分裂事件中发挥作用 (5),用 siRNA 使得 Aurora B 静默表达可导致组蛋白 H3 磷酸化、染色体对齐/隔离异常、生存素定位改变 (6)。
