方法 | Cell proliferation assay The effect of resveratrol on the cell growth of CNE-1 and CNE-2Z cells was examined using the MTT assay. Cancer cells were plated in 96-well plates at 104 cells/well. Following pretreatment with different concentrations of resveratrol for 24 or 48 h, viable cells were determined using the MTT Assay kit (Chemicon, Temecula, CA, USA) according to the manufacturer’s protocol. At indicated time-points, 20 μl of MTT solution (5 mg/ml in PBS) were added into each well and incubated for 4 h. Then, 100 μl of DMSO was added to dissolve the crystals. The plate was then left to stand for 10 min at room temperature and the corresponding absorbance was measured at 490 nm. Each experiment was performed in triplicate. |
