CC -!- FUNCTION: Secreted effector that hijacks host cell SUMOylation during
CC A.phagocytophilum infection and is important for the pathogen's
CC intracellular survival. {ECO:0000269|PubMed:25308709}.
CC -!- SUBCELLULAR LOCATION: Secreted {ECO:0000269|PubMed:20212090,
CC ECO:0000269|PubMed:25308709}. Host membrane
CC {ECO:0000269|PubMed:20212090}; Multi-pass membrane protein
CC {ECO:0000305|PubMed:20212090}. Host cytoplasm, host cytosol
CC {ECO:0000269|PubMed:25308709}. Note=Localizes to the A.phagocytophilum-
CC occupied vacuolar membrane (AVM) (PubMed:20212090). Potentially
CC traverses the AVM 3 or fewer times by means of the 3 bilobed
CC hydrophobic alpha-helical domains that are predicted to be in each
CC direct repeat (PubMed:20212090). Colocalizes with SUMO2/3 on the AVM
CC and in the host cytosol throughout the infection cycle
CC (PubMed:25308709). Only late during the infection cycle AmpA
CC colocalizes with SUMO1, which terminally caps poly-SUMO2/3 chains
CC (PubMed:25308709). Is probably not secreted via the type IV secretion
CC system (T4SS) (PubMed:20212090). {ECO:0000269|PubMed:20212090,
CC ECO:0000269|PubMed:25308709}.
CC -!- INDUCTION: Is induced within the initial hours of A.phagocytophilum
CC infection and is strongly expressed during the bacterium's
CC intracellular replication phase. {ECO:0000269|PubMed:20212090}.
CC -!- PTM: Polysumoylated during infection on at least two lysine residues,
CC in the N- and C-terminal section. SUMO2/3 modification of AmpA
CC throughout the infection cycle is likely critical for bacterial
CC intracellular survival, while terminal SUMO1 conjugation of AmpA may
CC promote a late-stage infection cycle event. Only a small portion of the
CC available AmpA pool is actually SUMOylated at any given time.
CC {ECO:0000269|PubMed:25308709}.
CC -!- MISCELLANEOUS: Pharmacologically inhibiting sumoylation in infected
CC cells significantly reduces the bacterial load.
CC {ECO:0000269|PubMed:25308709}.
