CC -!- FUNCTION: [Lanthionine-containing peptide SapB precursor RamS]: Stably
CC accumulated precursor of SapB (PubMed:22486809).
CC {ECO:0000269|PubMed:22486809}.
CC -!- FUNCTION: [Lanthionine-containing peptide SapB]: Lanthionine-containing
CC peptide devoid of antibiotic properties (PubMed:15277670). A surface
CC active peptide involved in the efficient formation of aerial mycelium
CC when cells are grown in rich media. Has an overlapping function with
CC the surface-active chaplin proteins; chaplins are essential on minimal
CC medium while on rich medium both chaplins and SapB are required for
CC efficient aerial hyphae formation (PubMed:17462011). Required under
CC conditions of high osmolarity where it may change the physical
CC properties of the chaplin layer to allow hyphae to grow into air
CC (Probable). Suggested to self-assemble at air-water interfaces, thus
CC providing a film of surfactant through which nascent aerial hyphae can
CC emerge; the aerial hyphae differentiate further into spores
CC (PubMed:2032288). Application to bald mutants (bld, unable to make
CC aerial hyphae) restores hyphae growth (PubMed:2032288, PubMed:9822824).
CC Application to chaplin negative mutants as well as ramC-ramS-ramA-ramB
CC and ramR deletions also restores aerial hyphae growth and sporulation
CC (PubMed:17462011). Reduces surface tension of water from 72 to 30
CC mJ/m(2) (PubMed:9822824). {ECO:0000269|PubMed:15277670,
CC ECO:0000269|PubMed:17462011, ECO:0000269|PubMed:2032288,
CC ECO:0000269|PubMed:9822824, ECO:0000305|PubMed:22309453}.
CC -!- SUBCELLULAR LOCATION: [Lanthionine-containing peptide SapB precursor
CC RamS]: Cell membrane {ECO:0000269|PubMed:22486809}. Note=RamS precursor
CC protein (detected by an anti-propeptide antibody and by a C-terminal
CC His-tag) is associated with the cell membrane in a non ramR-dependent
CC fashion (PubMed:22486809). {ECO:0000269|PubMed:22486809}.
CC -!- SUBCELLULAR LOCATION: [Lanthionine-containing peptide SapB]: Secreted
CC {ECO:0000269|PubMed:2032288, ECO:0000269|PubMed:22309453}. Spore wall
CC {ECO:0000269|PubMed:22309453, ECO:0000269|PubMed:2450872,
CC ECO:0000269|PubMed:9822824, ECO:0000305|PubMed:17462011}. Note=Present
CC in a zone around colonies undergoing aerial hyphae formation and around
CC spores (PubMed:2032288). Expression levels are media-dependent; well
CC expressed in spores grown on rich R5, about 4-fold less on rich MS and
CC less again on minimal mannitol media. Secreted in rich R5 but not in
CC minimal mannitol media (PubMed:22309453). {ECO:0000269|PubMed:2032288,
CC ECO:0000269|PubMed:22309453}.
CC -!- DEVELOPMENTAL STAGE: Starts to be expressed as aerial mycelium forms on
CC rich solid medium after 3 days growth and in spores; only forms during
CC growth on solid medium (PubMed:2450872, PubMed:2032288). Strongly
CC expressed in spores (PubMed:2450872, PubMed:9822824). Most strongly
CC expressed in spores from rich R5 media, less well in rich MS and even
CC less well on minimal mannitol media (at protein level)
CC (PubMed:22309453). {ECO:0000269|PubMed:2032288,
CC ECO:0000269|PubMed:22309453, ECO:0000269|PubMed:2450872,
CC ECO:0000269|PubMed:9822824}.
CC -!- INDUCTION: The RamS precursor protein (detected by an anti-propeptide
CC antibody and by a C-terminal His-tag) is detected from at least 16
CC hours post-germination on both rich and minimal media and is detected
CC for at least 52 hours (at protein level) (PubMed:22486809). The mature
CC lanthionine-containing peptide SapB is expressed by 3 days growth on
CC rich medium, expression increases slightly over 7 days (at protein
CC level). Expression requires ramR, not expressed in minimal medium (at
CC protein level) (PubMed:12453210). Not synthesized in bld mutants
CC (blocked in aerial hyphae formation), synthesized in whi mutants (spore
CC formation is prevented) (at protein level) (PubMed:2032288). SapB is
CC expressed on rich but not minimal medium (at protein level)
CC (PubMed:2032288, PubMed:17462011). However it is expressed after 6 days
CC growth on minimal medium in a chaplin-negative strain (at protein
CC level) (PubMed:17462011). Induced by growth on osmolytes; slight
CC induction by 10% sucrose, more strongly induced by 0.5 M KCl (at
CC protein level) (PubMed:22309453). Probably part of the ramC-ramS-ramA-
CC ramB operon (PubMed:12100547, PubMed:12453210). Also has its own, ramR-
CC independent promoter. On both rich and minimal medium transcription is
CC constitutive, rising to a plateau about 24 hours post-germination on
CC all media; unaffected by ramR disruption (PubMed:22486809).
CC {ECO:0000269|PubMed:12100547, ECO:0000269|PubMed:12453210,
CC ECO:0000269|PubMed:17462011, ECO:0000269|PubMed:2032288,
CC ECO:0000269|PubMed:22309453, ECO:0000269|PubMed:22486809}.
CC -!- PTM: Maturation involves the enzymatic conversion of Ser into
CC dehydrated AA and the formation of thioether bonds with cysteine,
CC probably by RamC (PubMed:15277670). This is followed by membrane
CC translocation and cleavage of the modified precursor (Probable). The
CC RamS precursor protein (detected by an anti-propeptide antibody and by
CC a C-terminal His-tag) is detected from at least 16 hours post-
CC germination; its apparent molecular weight decreases starting from
CC about 34 hours, when its probable modifying enzyme ramC is transcribed.
CC Surfactin, a B.subtilis cyclic lipopeptide antibiotic which prevents
CC aerial hyphae formation in S.coelicolor, decreases localization of RamS
CC precursor protein to the cell membrane, suggesting that processing only
CC occurs at the cell membrane (PubMed:22486809).
CC {ECO:0000269|PubMed:15277670, ECO:0000269|PubMed:22486809,
CC ECO:0000305|PubMed:15277670}.
CC -!- MASS SPECTROMETRY: [Lanthionine-containing peptide SapB]: Mass=2027;
CC Method=FAB; Evidence={ECO:0000269|PubMed:2032288};
CC -!- MASS SPECTROMETRY: [Lanthionine-containing peptide SapB]:
CC Mass=2026.9797; Mass_error=0.0109; Method=MALDI;
CC Evidence={ECO:0000269|PubMed:15277670};
CC -!- MASS SPECTROMETRY: [Lanthionine-containing peptide SapB]: Mass=2027.79;
CC Method=MALDI; Note=On rich R5 medium.;
CC Evidence={ECO:0000269|PubMed:22309453};
CC -!- MASS SPECTROMETRY: [Lanthionine-containing peptide SapB]: Mass=2027.50;
CC Method=MALDI; Note=On rich MS medium.;
CC Evidence={ECO:0000269|PubMed:22309453};
CC -!- DISRUPTION PHENOTYPE: Single deletion of ramS leads to a severe delay
CC in aerial hyphae growth (PubMed:12453210). Deletion of the ramC-ramS-
CC ramA-ramB operon on rich medium leads to an initially bald (no aerial
CC hyphae) phenotype; after 4 days develops a substantial aerial mycelium.
CC Wild-type mycelium on minimal medium (PubMed:17462011,
CC PubMed:22309453). No expression of SapB, normal expression of chaplins.
CC A complete chaplin-negative plus ram-negative strain (deletion of ramR
CC or the ramC-ramS-ramA-ramB operon) leads to the complete loss of robust
CC aerial hyphae (PubMed:17462011). {ECO:0000269|PubMed:12453210,
CC ECO:0000269|PubMed:17462011, ECO:0000269|PubMed:22309453}.
CC -!- SIMILARITY: Belongs to the lanthionine-containing morphogen family.
CC {ECO:0000305}.
