CC -!- FUNCTION: Toxic component of a type II toxin-antitoxin (TA) system.
CC Upon overexpression inhibits cell growth and colony formation. Its
CC toxic effect is neutralized by coexpression with cognate antitoxin
CC MazE. In M.smegmatis 3 TA systems (VapB-VapC, MazE-MazF and Phd-Doc)
CC may be involved in monitoring the nutritional supply and physiological
CC state of the cell, linking catabolic with anabolic reactions
CC (PubMed:22199354). Probably an endoribonuclease (By similarity).
CC {ECO:0000250|UniProtKB:P9WIH9, ECO:0000269|PubMed:22199354}.
CC -!- SUBUNIT: Forms a complex with cognate antitoxin MazE.
CC {ECO:0000250|UniProtKB:P9WIH9}.
CC -!- INDUCTION: Constitutively expressed during exponential growth and early
CC stationary phase, it decreases in late stationary phase; part of the
CC mazE-mazF operon. Negatively autoregulated by one or both of MazE-MazF.
CC {ECO:0000269|PubMed:22199354}.
CC -!- DISRUPTION PHENOTYPE: The mazE-mazF operon is not essential. A triple
CC TA mutant (missing vapB-vapC, mazE-mazF and phd-doc TA systems)
CC survives antibiotic challenge, suggesting the TA systems are not
CC required to generate drug-resistant cells. However the mutant is more
CC sensitive to oxidative and heat stress, and does not survive long term
CC starvation during aerobic growth on complex medium. There is a
CC difference in the level of branched-chain amino acids, which may play a
CC role in monitoring the nutritional supply and physiological state of
CC the cell. {ECO:0000269|PubMed:22199354}.
CC -!- SIMILARITY: Belongs to the PemK/MazF family. {ECO:0000305}.
