CC -!- FUNCTION: Acts as an orphan toxin which is important for maintaining
CC cell fitness during stress related to the stringent response (decreased
CC amino acid, purine and thymidine availability). Overexpression inhibits
CC cell growth and increases the formation of persister cells. Causes
CC 99.9% of cells to undergo bacterial lysis within 2 hours after
CC induction; nucleoids condense, the cytoplasm seems empty and the
CC periplasmic space enlarges. The intracellular ATP level decreases about
CC 27-fold suggesting the membrane potential may be disrupted.
CC {ECO:0000269|PubMed:25643179}.
CC -!- SUBCELLULAR LOCATION: Cell inner membrane
CC {ECO:0000305|PubMed:25643179}; Multi-pass membrane protein
CC {ECO:0000305}.
CC -!- INDUCTION: By conditions that induce the stringent response such as
CC increased expression of RelA, heat shock at 43 degres Celsius, probably
CC under control of sigma factor E and/or H (rpoE, rpoH), or growth in the
CC presence of trimethoprim (TMP), mupirocin or serine hydroxamate.
CC {ECO:0000269|PubMed:25643179}.
CC -!- DISRUPTION PHENOTYPE: Strongly represses cell swarming but no effect on
CC cell swimming (PubMed:17122336). Increased metabolic activity in the
CC presence of TMP, an inhibitor of dihydrofolate reductase (folA) and
CC sulfamethoxazole, an inhibitor of dihydropteroate synthetase (folC).
CC Cells grow faster than wild-type in the presence of TMP.
CC {ECO:0000269|PubMed:17122336, ECO:0000269|PubMed:25643179}.
CC -!- MISCELLANEOUS: Although this protein has high homology to GhoT, the
CC toxic component of a type IV toxin-antitoxin system, no antitoxin has
CC been found. GhoS (the antitoxin for GhoT), YdcY (the neighboring gene),
CC 5'- and 3'-UTRs as well as approximately 620 bp lengths of the BW25113
CC genome DNA were tested. {ECO:0000269|PubMed:25643179}.
CC -!- SIMILARITY: Belongs to the GhoT/OrtT toxin family. {ECO:0000305}.
